Abstract: The use of low-level laser (LLL) has been described as a method that can promote improvement in bone repair. However, there are only few data in the literature about the precise molecular biological processes resulting from the effect of therapy with LLL in the repair of critical-size defects. The aim of this study was to evaluate the modulation of bone extracellular matrix in bone defects (Ө = 8mm) created on rat calvaria. The bone healing was analysed after 2, 4 and 8 weeks in two groups: control and irradiated (laser gallium-aliminum arsenide / GaAlAs, 830 nm, energy dose of 2.5 J/cm2 and power of 50 mW in a total of seven applications, one every 48 hours). Immunohistochemistry was performed to determine the immunoreactivity of osteocalcin and subsequently proceeded histomorphometric analysis and statistical analysis. Comparison between irradiated group and control did not present intergroups significance in the tested periods, although the group of GaAlAs laser had a greater statistical difference 8 weeks with other experimental times and higher rate of deposition of osteocalcin on the total volume of bone, interpreted as a more mature newly formed tissue. In accord to the expression peak of osteocalcin have been delayed, the profile of other bone biomarkers deserves to be further investigated to understand the process of early biostimulation promoted by GaAlAs laser.
